Our will is to make innovative chemical tools available to the research community worldwide. We are very proud to have so far commercialized three types of innovative products: membrane tension probes, turn-on probes for bioimaging and a new light-activated lipid molecule under exclusive distributors (Spirochome and Avanti). These tools showcase the performance of the NCCR Chemical Biology.
Membrane tension probes
Fluorescent membrane tension probe (Flipper-TR ® )
Flipper-TR ® (published under the name FliptR) is the first live cell fluorescent membrane tension probe developed for the field of mechanobiology. The fluorescence lifetime ofFlipper-TR ® is strongly dependent on the membrane tension, therefore through FLIM imaging it is possible to measure precisely the spatio-temporal distribution of tension in membranes. This chemical tool opens up a whole new field by giving access to real time membrane tension measurements in live cells and artificial membranes.
Mito-caged sphingosine is a new light-activated lipid molecule synthesized as an inactive molecule that would only localize to mitochondria and which can be activated with a flash of light. In human cells, activated mito-caged sphingosine is converted to sphingosine 1 phosphate within the mitochondria, before being exported rapidly to another compartment. The new compound represents a useful tool for many researchers studying both metabolism and signaling as well as those seeking a better understanding of mitochondria biology.
Probes for bioimaging
1. SiR actin kit
SiR-actin is a fluoregenic, cell permeable and highly specific probe for F-actin. Sir-actin is based on the F-actin binding natural product jasplakinolide. Sir-actin stains endogenous F-actin without the need for genetic manipulation or overexpression. Its emission in the far red minimizes phototoxicity and sample autofluorescence. SiR-actin is compatible with GFP and/or m-cherry fluorescent proteins. It can be imaged with standard Cy5 filtersets. SiR-actin can be used for widefield, confocal, SIM or STED imaging in living cells and tissue.
2. SiR-tubulin kit
SiR-tubulin is a fluorogenic, cell permeable and highly specific probe for microtubules. SiR-tubulin is based on the microtubule binding drug Docetaxel. SiR-tubulin is fluorogenic, cell permeable and highly specific for microtubules. Sir-tubulin stains endogenous microtubules without the need for genetic manipulation or overexpression. Its emission in the far red minimizes phototoxicity and sample autofluorescence. SiR-tubulin is compatible with GFP and/or m-cherry fluorescent proteins. It can be imaged with standard Cy5 filtersets. SiR-tubulin can be used for widefield, confocal, SIM or STED imaging in living cells and tissue.
